چکیده انگلیسی مقاله Background and Objective: Toll-like receptors (TLRs) have been discovered as the most important receptors in innate immunity. One of the most important TLRs is TLR4, the key receptor for the LPS component of gram-negative bacteria. Two polymorphisms, D299G (rs4986790) and T399I (rs4986791), in TLR4 gene are associated with a decreased response to LPS. This study was done to estimate the expression of different polymorphisms of TLR4 gene in colorectal cancer cell line by flowcytometery. Materials and Methods: In this laboratory study, the HCT116 cells were transfected with plasmids containing different variants of TLR4 gene including; Flag-tagged-TLR4 wild type, flag-tagged D299G and T399I Using TurboFect transfection reagent. Transfection efficiency was evaluated by GFP plasmid. Expression of different variants of TLR4 was assessed in transfected cells by flowcytometery. Data were analyzed using SPSS-11.5 and chi-square test. Results: TLR4 was detected on HT29 and CaCo2 cell lines at low levels. HCT116 cells did not express detectable amounts of TLR4 by flowcytometery prior to transfection. Gene transfer efficiency for GFP plasmid was about 80% in HCT116 cells by flowcytometery and microscopic analysis. TLR4 expression and LPS responsiveness significantly was higher in HCT116 cells which were transfected with wild type TLR4 gene compared to non-transfected and mutant transfected cells (P<0.05). Conclusion: Lower expression of TLR4 on cells with mutant TLR4 showed that these polymorphisms affect on expression patterns of TLR4 on colon cancer cells.
چکیده فارسی مقاله زمينه و هدف : گيرنده‌های شبه Toll (TLRs) به عنوان مهم‌ترين گيرنده‌ها در ايمنی ذاتی شناخته شده‌اند. گيرنده TLR4 به عنوان گيرنده اصلی در شناسايی ليپوپلی‌ساکاريد (LPS) باکتری‌های گرم منفی عمل می‌کند. دو پلی‌مورفيسم rs4986790) D299G (SNP و (SNP rs4986791) T399I در ژن اين گيرنده موجب کاهش پاسخ‌دهی به LPS می‌شود. اين مطالعه به منظور تعيين ميزان بيان پلی‌مورفيسم‌های مختلف ژن TLR4 در رده سلولی سرطان کولورکتال HT29 ، HCT116 و CaCo2 به روش فلوسايتومتری انجام شد. روش بررسی : در اين مطالعه آزمايشگاهی انتقال پلاسميد حاوی ژن واريته‌های مختلف شامل Flag-tagged-TLR4وحشی، Flag-tagged D299G و Flag-tagged T399I موتان به سلول‌های HCT116 توسط معرف Turbofect صورت گرفت. راندمان انتقال ژن توسط پلاسميد GFP ارزيابی شد. ميزان بيان واريته‌های مختلف TLR4 در سلول‌های دريافت کننده ژن‌ها از طريق فلوسايتومتری بررسی شد. داده‌ها با استفاده از نرم‌افزار آماری SPSS-11.5 و آزمون Chi-Square تجزيه و تحليل شدند. يافته‌ها : رده‌های سلولی HT29 و CaCo2گيرنده TLR4 را در سطح پايينی بيان کردند. در حالی که سلول‌های HCT116 قبل از انتقال پلاسميد، ژن TLR4 را در سطح قابل رديابی به روش فلوسايتومتری بيان نکردند. راندمان انتقال ژن GFP به داخل سلول‌های HCT116 به‌وسيله آناليز ميکروسکوپی و فلوسايتومتری حدود 80 درصد اندازه‌گيری شد. براين اساس ميزان بيان TLR4و پاسخ‌دهی به LPS در سلول‌هايی که ژن واريته وحشی TLR4 را دريافت کردند؛ به‌طور معنی‌داری بيشتر از سلول‌هايی بود که ژن واريته موتان را دريافت کردند (P<0.05). نتيجه‌گيری : سطح پايين بيان TLR4 روی سلول‌های دارای نوع موتان گيرنده نشان داد که اين پلی‌مورفيسم‌ها روی بيان گيرنده در سلول‌های سرطان کولون اثر دارند.
